Thalidomide in severe orogenital ulceration.

J S Jenkins, R J Powell, B R Allen

    Lancet (London, England)
    |December 22, 1984

    Summary

Thalidomide effectively treats severe orogenital ulceration (OGU), leading to complete healing in most patients. Glutethimide showed no therapeutic benefit, and no adverse neurological effects were observed with thalidomide.

    Area of Science:

    • Immunology
    • Dermatology
    • Pharmacology

    Background:

    • Severe orogenital ulceration (OGU) presents a significant clinical challenge.
    • Limited effective therapeutic options exist for debilitating OGU.
    • Thalidomide has shown promise in other inflammatory conditions.

    Purpose of the Study:

    • To evaluate the efficacy and safety of thalidomide in patients with severe OGU.
    • To compare thalidomide treatment against a placebo (glutethimide).

    Main Methods:

    • A study involving 15 patients with severe OGU.
    • Four patients received thalidomide in a double-blind, placebo-controlled trial using glutethimide.
    • Eleven patients received open-label thalidomide treatment.

    Related Experiment Videos

    Main Results:

    • Complete ulcer resolution in 14 out of 15 patients.
    • Significant improvement in the remaining patient.
    • No development of peripheral neuropathies during thalidomide treatment.
    • Patients did not respond to glutethimide placebo.

    Conclusions:

    • Thalidomide is a highly effective treatment for severe orogenital ulceration.
    • Thalidomide demonstrates a favorable safety profile regarding neuropathy in this context.
    • Contraceptive measures are essential during thalidomide therapy due to potential teratogenicity.
