    Summary

    This study investigated a viral cause for Behçet's syndrome by examining patient lymphocytes. Results showed impaired herpes simplex virus (HSV) growth and increased chromosomal abnormalities, supporting a potential viral etiology.

    Background:

    • Behçet's syndrome is a complex inflammatory disorder with an unknown etiology.
    • A viral cause for Behçet's syndrome has been hypothesized but not definitively proven.

    Purpose of the Study:

    • To indirectly test the viral hypothesis for Behçet's syndrome.
    • To evaluate lymphocytes from Behçet's patients for markers of persistent viral infection.

    Main Methods:

    • Assessed lymphocytes from 86 Behçet's patients and controls.
    • Measured chromosomal abnormalities in lymphocytes.
    • Evaluated the ability of herpes simplex virus type I (HSV) to replicate in PHA-stimulated lymphocyte cultures.

  • HSV replication was impaired in 37% of Behçet's patients' lymphocytes (57% excluding those on steroids/cytotoxic drugs).
  • Chromosomal abnormalities were found in 16 of 38 examined patients, versus 1 of 17 controls.
  • Chromosomal damage was linked to impaired HSV replication and inversely to steroid treatment.
  • Conclusions:

    • Findings are consistent with a viral etiology for Behçet's syndrome.
    • Further research is needed to exclude other potential causes.
