    Area of Science:

    • Molecular Biology
    • Biochemistry
    • Genetics

    Background:

    • U4 RNA is a capped nuclear small nuclear RNA (snRNA) found in ribonucleoprotein particles (RNPs).
    • snRNPs, including U4 RNP, are implicated in heterogeneous nuclear RNA (hnRNA) processing and messenger RNA (mRNA) transport.
    • The conserved protein components of different snRNPs suggest RNA moieties dictate specific functions.

    Purpose of the Study:

    • To determine the complete nucleotide sequence of U4 RNA.
    • To elucidate the functional role of U4 RNA within U4 RNP particles.
    • To provide sequence data for understanding snRNP-mediated cellular processes.

    Main Methods:

    • Isolation and purification of U4 RNA from ribonucleoprotein particles.
    • Determination of the nucleotide sequence of U4 RNA using established biochemical methods.
    • Analysis of modified nucleotides and sequence microheterogeneity.

    Main Results:

    • The complete nucleotide sequence of U4 RNA was established.
    • U4 RNA contains modified nucleotides, including N2,2-dimethylguanosine (Am) at position 63 and N6-methyladenosine (m6A) at position 98.
    • A-C microheterogeneity was observed at position 97 of the U4 RNA sequence.

    Conclusions:

    • The determined nucleotide sequence of U4 RNA provides a critical resource for understanding its structure and function.
    • The presence of specific modified nucleotides and microheterogeneity may play roles in U4 RNP assembly and function.
    • This sequence data is essential for further investigations into the mechanisms of hnRNA processing and mRNA transport involving U4 snRNPs.
