    Summary

    Background:

    • Aplastic anemia is a rare but serious blood disorder.
    • Bleomycin is a chemotherapeutic agent known to induce DNA strand breaks.

    Purpose of the Study:

    • To investigate DNA integrity and bleomycin sensitivity in patients with aplastic anemia.
    • To explore potential molecular mechanisms underlying aplastic anemia.

    Main Methods:

    • Ultracentrifugation on alkaline sucrose density gradients was used to analyze DNA.
    • DNA strand breaks were quantified before and after bleomycin exposure.
    • Lymphocyte sensitivity to bleomycin in tissue culture was assessed.

    Main Results:

    • DNA from aplastic anemia patients exhibited more strand breaks than controls prior to bleomycin exposure.
    • 10 out of 14 aplastic anemia patients showed an abnormal number of DNA strand breaks after bleomycin exposure.
    • This molecular finding correlated with increased sensitivity of proliferating lymphocytes to bleomycin in vitro.
    • Two relatives of patients also displayed similar DNA abnormalities and bleomycin sensitivity.

    Conclusions:

    • Some patients with aplastic anemia may have underlying abnormalities in DNA structure or repair.
    • Bleomycin sensitivity serves as a potential molecular marker for aplastic anemia.
    • These findings may aid in understanding the pathogenesis of aplastic anemia.
