    Summary

    Estrogen receptor was found in human benign prostatic hyperplasia (BPH) tissue, supporting the theory that estrogens play a role in BPH development. This finding was consistent across normal and cancerous prostate tissues as well.

    Background:

    • Estrogens are implicated as a key factor in benign prostatic hyperplasia (BPH) pathogenesis.
    • The presence of estrogen receptors (ER) in BPH tissue would substantiate this hypothesis.

    Purpose of the Study:

    • To investigate the presence and characteristics of cytosolic estrogen receptor in human benign prostatic hyperplasia (BPH) tissue.
    • To compare ER concentrations with those in normal and cancerous prostate tissues.
    • To measure androgen receptor (AR) and progesterone receptor (PR) concentrations in these tissues.

    Main Methods:

    • Cytosolic estrogen receptor was assayed in human BPH, normal, and cancerous prostate tissues using a hydroxylapatite assay with sodium molybdate as a receptor stabilizer.
    • Sucrose gradient analysis and steroid specificity studies were performed to characterize the estrogen receptor.
    • Androgen receptor and progesterone receptor concentrations were also measured.

    Main Results:

    • Estrogen receptor was detected in 8 of 15 (53%) benign prostatic hyperplasia specimens at a mean concentration of 9.2 fmol/mg protein.
    • The estrogen receptor in BPH tissue exhibited sedimentation characteristics of 8S and demonstrated estrogen-specific binding.
    • Estrogen receptor was also found in normal (3/3) and malignant (4/6) prostate tissues, with all tissues positive for androgen receptor.

    Conclusions:

    • The presence of estrogen receptor in human benign prostatic hyperplasia supports the role of circulating estrogens in the pathogenesis of this condition.
    • Estrogen receptor is present in normal and malignant prostate tissues, indicating a potential broader role in prostate physiology and pathology.
