    Summary

    Polyamine biosynthesis is significantly elevated in psoriatic lesions, with increased spermidine levels also noted in unaffected psoriatic skin. Arginase activity is also enhanced in psoriatic skin, particularly in the epidermis.

    Background:

    • Psoriasis is a chronic inflammatory skin condition.
    • Polyamines and arginase play roles in cell proliferation and inflammation.
    • Altered polyamine metabolism is implicated in various skin diseases.

    Purpose of the Study:

    • To investigate polyamine biosynthesis and arginase activity in psoriatic skin.
    • To compare these parameters in lesional and uninvolved psoriatic skin with healthy controls.

    Main Methods:

    • Analysis of polyamine levels and enzyme activities (ornithine decarboxylase, S-adenosyl-L-methionine decarboxylase, arginase) in skin specimens.
    • Specimens included keratotome strips and suction blister roofs from psoriatic patients and controls.

    Main Results:

    • Uninvolved psoriatic skin showed a slight increase in spermidine and the spermidine/spermine ratio compared to controls.
    • Psoriatic lesions exhibited significantly elevated levels of putrescine, spermidine, and spermine.
    • Activities of polyamine biosynthetic enzymes and arginase were significantly increased in psoriatic lesions.
    • Arginase activity increase was more pronounced in pure epidermis (suction blister roofs).

    Conclusions:

    • Polyamine biosynthesis is significantly upregulated in active psoriatic lesions.
    • Subtle alterations in polyamine levels are present even in uninvolved psoriatic skin.
    • Arginase activity is also enhanced in psoriatic skin, suggesting its involvement in the disease pathogenesis.
