Bacillus subtilis requires a "stringent" Shine-Dalgarno region for gene expression.

    Summary

Bacterial expression of leukocyte interferon-A is sensitive to Shine-Dalgarno sequences. Bacillus subtilis showed higher sensitivity than Escherichia coli, requiring greater homology to ribosomal RNA for efficient interferon production.

    Area of Science:

    • Molecular Biology
    • Microbial Genetics
    • Protein Expression

    Background:

    • The Shine-Dalgarno sequence is crucial for initiating protein synthesis in prokaryotes.
    • Optimizing gene expression in different bacterial hosts is essential for biotechnology.

    Purpose of the Study:

    • To investigate the impact of Shine-Dalgarno sequence variations on leukocyte interferon-A expression.
    • To compare the efficiency of interferon expression in Bacillus subtilis and Escherichia coli.

    Main Methods:

    • Construction of a plasmid series with varied Shine-Dalgarno sequences.
    • Expression analysis of leukocyte interferon-A in B. subtilis and E. coli.

    Main Results:

    • Interferon expression in B. subtilis was highly sensitive to Shine-Dalgarno sequence changes.

    Related Experiment Videos

  • E. coli showed less sensitivity to Shine-Dalgarno sequence variations.
  • Optimal expression in B. subtilis required increased homology to the 3' end of 16S ribosomal RNA.
  • Conclusions:

    • Shine-Dalgarno sequence optimization is critical for efficient interferon production in B. subtilis.
    • Bacterial host choice significantly influences gene expression based on Shine-Dalgarno sequence recognition.
