    Summary

    Bone marrow contains immature natural killer (NK) cell precursors that can be cultured to generate significant cytotoxic activity. These NK cells, identified as Nk-1+ and Qa-5+, demonstrate potent anti-tumor potential.

    Background:

    • Natural killer (NK) cells are crucial for innate immunity.
    • Immature NK cell precursors reside in the bone marrow.
    • Nk-1 antigen is a marker for NK cells.

    Purpose of the Study:

    • To investigate the presence and differentiation of NK cell precursors in bone marrow.
    • To characterize the cytotoxic activity of cultured bone marrow-derived NK cells.
    • To compare bone marrow-derived NK cell differentiation with spleen-derived NK cells.

    Main Methods:

    • Detection of Nk-1+ cells in bone marrow using anti-Nk-1 antiserum.
    • Culture of bone marrow cells in concanavalin A (Con A)-conditioned medium to induce differentiation.
    • Assessment of cytotoxic activity against YAC-1 cells.
    • Phenotypic analysis of generated cytotoxic cells (Nk-1+, Qa-5+).

    Main Results:

    • Bone marrow harbors Nk-1+ cells with low lytic activity, slightly enhanced by interferon (IF).

    • Bone marrow contains immature NK cell precursors capable of differentiation into cytotoxic NK cells.
    • Con A-conditioned medium is effective in inducing NK cell differentiation and cytotoxic function from bone marrow precursors.
    • Bone marrow-derived NK cells show robust cytotoxic potential, even in aged mice.
