    Background:

    • Tricyclic antidepressants (TCAs) are commonly prescribed.
    • Optimizing dosing regimens is crucial for therapeutic efficacy.

    Purpose of the Study:

    • To evaluate the pharmacokinetic differences of a single 100-mg dose of nortriptyline administered in the morning versus the evening.

    Main Methods:

    • Ten healthy male subjects received a 100-mg dose of nortriptyline at 9 A.M. and 9 P.M. on separate occasions.
    • Plasma concentrations of nortriptyline and its metabolite 10-hydroxynortriptyline were analyzed.

    Main Results:

    • Minimal differences in pharmacokinetic parameters were observed between morning and evening dosing.
    • High plasma concentrations of 10-hydroxynortriptyline indicated significant first-pass metabolism.

    Conclusions:

    • Evening administration of single daily doses of nortriptyline is unlikely to offer significant pharmacokinetic advantages over morning dosing.
    • First-pass metabolism plays a substantial role in nortriptyline's disposition.
