Substance P concentrations in the rat tubero-hypophyseal system vary by region. Dehydration and sodium loading significantly reduced neural lobe substance P levels, indicating its role in neurohormone secretion.

    Area of Science:

    • Neuroendocrinology
    • Neuropeptide Research

    Background:

    • The tubero-hypophyseal system regulates hormone secretion.
    • Substance P is a neuropeptide found in the central nervous system.

    Purpose of the Study:

    • To quantify Substance P levels in different rat pituitary regions.
    • To investigate the effect of capsaicin and dehydration/sodium loading on Substance P concentrations.

    Main Methods:

    • Radioimmunoassay to measure immunoreactive Substance P (I-SP).
    • Experimental manipulation involving early-life capsaicin treatment.
    • Induction of altered neurohormone secretion via dehydration and sodium loading.

    Main Results:

    • Baseline I-SP concentrations were highest in the medial basal hypothalamus (MBH), followed by the anterior lobe (AL) and neural lobe (NL).

    Related Experiment Videos

  • Capsaicin treatment did not alter I-SP levels in any pituitary region.
  • Dehydration and sodium loading led to a 70% decrease in NL I-SP, with no changes in AL or MBH.
  • Conclusions:

    • Substance P distribution in the rat tubero-hypophyseal system is region-specific.
    • Neural lobe Substance P levels are sensitive to changes in neurohormone secretion, suggesting a role in regulating this process.
