    Activated rabbit macrophages (AMs) show altered IgG and complement (C) receptors, with increased IgG-mediated phagocytosis indicating activation. Complement-mediated phagocytosis remains low in rabbit macrophages compared to mice.

    Background:

    • Immune cell receptors, such as those for IgG and complement (C), can change with activation.
    • Alveolar macrophage (AM) populations, constantly exposed to particulates, might resist receptor modulation upon activation.

    Purpose of the Study:

    • To compare normal and activated rabbit alveolar macrophages (AMs) and peritoneal macrophages (PMs).
    • To investigate the impact of activation on IgG and C receptor function in rabbit macrophages.

    Main Methods:

    • Rabbit AMs and PMs were activated using intravenous BCG or intratracheal Micropolyspora faeni.
    • Rosette formation and phagocytosis assays were performed using sheep red blood cells (SRBC) coated with varying IgG or C concentrations.

    Main Results:

    • Activated AMs showed enhanced rosette formation and IgG receptor-mediated phagocytosis compared to normal AMs.
    • BCG-activated PMs exhibited increased IgG-mediated phagocytosis but not rosette formation.
    • Complement-mediated phagocytosis was infrequent in both normal and activated rabbit macrophages, and lower than in activated mouse PMs.

    Conclusions:

    • Rabbit AM IgG and C receptors are modifiable by activation events.
    • IgG-mediated phagocytosis serves as a reliable indicator of rabbit AM activation.
    • Rabbit macrophages display significantly lower complement-mediated phagocytosis than activated mouse macrophages.
