RecA protein--promoted lambda repressor cleavage: complementation between RecA441 and RecA430 proteins in vitro.

    Summary

The RecA441 protein, at low levels, can cleave the lambda repressor when RecA430 protein is present. Mixed RecA protein complexes on DNA enable prophage lambda induction in recA mutant lysogens.

    Area of Science:

    • Molecular Biology
    • Genetics
    • Biochemistry

    Background:

    • Prophage lambda induction is regulated by RecA protein activity.
    • Specific recA mutations (recA441, recA453) affect lambda induction.
    • RecA430 protein can restore induction but lacks repressor cleavage activity alone.

    Purpose of the Study:

    • To elucidate the mechanism of RecA430 protein complementation with RecA441 protein in lambda repressor cleavage.
    • To understand the role of mixed RecA protein-RecA430 and RecA441 multimers in DNA binding and cleavage.
    • To investigate the in vitro interaction of RecA441 and RecA430 proteins with single-stranded DNA and lambda repressor.

    Main Methods:

    • In vitro characterization of lambda repressor cleavage by mixed RecA441 and RecA430 proteins.

    Related Experiment Videos

  • Analysis of RecA protein-DNA complex formation and stability using various DNA substrates and nucleoside triphosphates.
  • Complementation assays in recA mutant lysogens.
  • Main Results:

    • RecA441 and RecA430 proteins form mixed multimers on single-stranded DNA in the presence of dATP.
    • RecA441 protein enhances RecA430 protein's DNA binding affinity, but RecA430 does not cleave the repressor.
    • Cooperative binding of RecA441 to DNA stabilizes complexes, enabling lambda repressor cleavage; SSB protein cannot substitute for RecA430.

    Conclusions:

    • A low basal level of RecA441 protein is sufficient for lambda repressor cleavage when RecA430 protein is present.
    • Formation of mixed RecA441-RecA430 multimers on single-stranded DNA is crucial for restoring prophage lambda induction.
    • RecA protein-DNA complex stability is key for efficient repressor cleavage and phage induction.
