    Purpose of the Study:

    • To definitively identify the cellular site of gastric acid secretion.
    • To visualize the dynamic changes in parietal cells during acid production.

    Main Methods:

    • Differential interference-contrast (Nomarski) microscopy to observe living gastric glands.
    • Histamine stimulation and weak base (aminopyrine) treatment to induce acid secretion.
    • Acridine orange fluorescence microscopy to track pH gradients and accumulation.
    • Electron microscopy to examine cellular ultrastructure.

    Main Results:

    • Histamine stimulation caused observable vacuolization in parietal cells, corresponding to swelling of secretory canaliculi.
