Polyoma virus minichromosomes: associated enzyme activities.

B B Gourlie, V Pigiet, C B Breaux

    Journal of Virology
    |June 1, 1981
    PubMed
    Summary

DNA polymerase alpha associates with replicating polyoma DNA, while DNA polymerase beta and topoisomerase I bind mature minichromosomes. RNase H shows partial association, and DNA polymerase gamma does not associate with these structures.

    Area of Science:

    • Molecular Biology
    • Virology
    • Biochemistry

    Background:

    • Polyoma minichromosomes are essential for viral DNA replication and packaging.
    • Understanding the enzymatic machinery associated with these structures is crucial for deciphering viral DNA metabolism.

    Purpose of the Study:

    • To investigate the association of specific DNA polymerases (alpha, beta, gamma), DNA topoisomerase I, and RNase H with polyoma minichromosomes.
    • To determine the localization of these enzymes relative to viral DNA replication intermediates and mature minichromosomes.

    Main Methods:

    • Isolation and glycerol gradient fractionation of polyoma minichromosomes.
    • Enzymatic assays for DNA polymerases alpha, beta, gamma, DNA topoisomerase I, and RNase H.
    • Quantitative electron microscopy to determine the number of polyoma DNA molecules and replicative intermediates.

    Related Experiment Videos

    Main Results:

    • DNA polymerase alpha activity cosedimented with polyoma replicative intermediate DNA.
    • DNA polymerase beta and DNA topoisomerase I activities were found associated with mature polyoma minichromosomes.
    • RNase H activity peaked slightly behind mature minichromosomes, while DNA polymerase gamma showed no association.

    Conclusions:

    • DNA polymerase alpha is involved in polyoma DNA replication.
    • DNA polymerase beta and DNA topoisomerase I likely play roles in the structure or maintenance of mature polyoma minichromosomes.
    • RNase H and DNA polymerase gamma have distinct roles, not directly associated with mature minichromosomes.
