    Summary

    The Sindbis virus causes ocular inflammation (uveoretinitis) in mice. The immune system clears the virus but also causes the eye disease, indicating immune-mediated pathology.

    Background:

    • Intraocular injection of Sindbis virus in BALB/c mice induces uveoretinitis.
    • Ocular disease manifests as iridocyclitis and retinitis with retinal dysplasia.

    Purpose of the Study:

    • To investigate the role of immunologic mechanisms in Sindbis virus-induced ocular disease.
    • To determine if the virus is directly cytopathogenic or if the host immune response mediates ocular pathology.

    Main Methods:

    • Adult BALB/c mice were infected intraocularly with Sindbis virus.
    • Immunosuppression was induced using cyclophosphamide post-infection.
    • Viral replication and inflammatory responses were monitored.

    Main Results:

    • Immunosuppression significantly reduced ocular inflammation, suggesting a non-cytopathogenic viral effect.
    • Virus replication was higher in immunosuppressed mice, but titers eventually decreased.
    • Inflammatory infiltrate comprised mainly lymphocytes and histiocytes.

    Conclusions:

    • The immune response responsible for viral clearance also mediates the ocular disease.
    • Sindbis virus-induced uveoretinitis is an immunologically driven condition, not solely due to viral cytopathogenicity.
