    Background:

    • Prazosin is an alpha-1 adrenergic antagonist.
    • Its effects on blood pressure and response to agonists can change with repeated dosing.

    Purpose of the Study:

    • To investigate the effects of oral prazosin on blood pressure.
    • To assess prazosin's antagonism of phenylephrine-induced blood pressure increases.
    • To evaluate changes in these effects after 3 days of dosing.

    Main Methods:

    • Six healthy subjects received oral prazosin.
    • Blood pressure was measured after the first dose and on day 3.
    • Phenylephrine dose-response curves were generated before and after prazosin administration.

    Main Results:

    • Prazosin lowered blood pressure more significantly after the first dose compared to day 3.
    • Tolerance developed to prazosin's blood pressure-lowering effects and phenylephrine's pressor response.
    • Phenylephrine's pressor response shifted right before prazosin on day 4, but left after prazosin dosing.

    Conclusions:

    • Repeated prazosin dosing leads to tolerance in blood pressure reduction and alpha-receptor antagonism.
    • This tolerance may be due to alpha-adrenoceptor desensitization.
    • Differential effects on agonist and antagonist activity were observed.
