    Summary

    This study investigated hormone receptor status in cystosarcoma phyllodes tumors. High-grade tumors showed significantly higher glucocorticoid receptor concentrations compared to low-grade tumors.

    Background:

    • Cystosarcoma phyllodes tumors are rare breast neoplasms with varying malignant potential.
    • Hormone receptor status is crucial for understanding tumor biology and guiding treatment in breast cancers.

    Purpose of the Study:

    • To analyze the expression and characteristics of estrogen receptors (ER), progesterone receptors (PR), and glucocorticoid receptors (GR) in cystosarcoma phyllodes tumors.
    • To correlate receptor status with tumor grade and patient menopausal status.

    Main Methods:

    • Histopathological classification of seven cystosarcoma phyllodes tumors into low-grade (5 cases) and high-grade (2 cases).
    • Quantification of ER, PR, and GR levels using biochemical assays (fmoles/mg protein).
    • Sucrose gradient centrifugation to determine receptor sedimentation characteristics.

    Main Results:

    • Low-grade tumors showed positivity for PR (100%), GR (60%), and ER (40%).
    • High-grade tumors exhibited higher GR concentrations (mean 275 fmoles/mg protein) compared to low-grade tumors (mean 27.2 fmoles/mg protein).
    • ER and PR were detected in one high-grade tumor, with slightly higher values than in low-grade tumors. ER and PR receptors were 4S-specific, while GR receptors sedimented faster (6-7S).

    Conclusions:

    • Hormone receptor expression, particularly GR, differs between low-grade and high-grade cystosarcoma phyllodes.
    • The distinct sedimentation properties of GR suggest unique molecular characteristics in these tumors.
    • Further research is warranted to elucidate the clinical implications of these findings.
