    Background:

    • Adrenocortical cells are crucial for hormone production.
    • Ascorbic acid (AA) plays a role in adrenal function.
    • Adrenocorticotrophin (ACTH) regulates adrenal cortex activity.

    Purpose of the Study:

    • To investigate the influence of ACTH on ascorbic acid (AA) accumulation in rat adrenocortical cells.
    • To differentiate between short-term and long-term effects of ACTH on AA uptake.

    Main Methods:

    • Primary cultures of rat adrenocortical cells were established.
    • Cells were treated with varying durations of ACTH.
    • Radiolabeled ascorbic acid (AA) uptake was measured and quantified per microgram of DNA.

    Main Results:

    • Long-term ACTH pretreatment (3 days) significantly increased AA accumulation (2-3 fold) compared to controls.
    • Acute ACTH administration concurrently with AA inhibited AA accumulation by approximately 30% in pretreated cells.
    • The acute inhibitory effect of ACTH on AA uptake was less pronounced in non-pretreated cells.

    Conclusions:

    • ACTH exhibits both long-term stimulatory and acute inhibitory effects on AA accumulation in adrenocortical cells.
    • The maintenance of the adrenal cortex's differentiated state is critical for maximal AA uptake and responsiveness to ACTH's inhibitory effects.
