    Summary

    Cyclic AMP-dependent protein kinase exists in the human placental chorion. Enzyme activity is highest in the first two trimesters, significantly decreasing in the third trimester.

    Background:

    • Limited research exists on cyclic AMP-dependent protein kinase (PKA) in the human placenta.
    • This study investigates PKA in the human placental chorion, characterizing its chemical properties and activity across gestation.

    Purpose of the Study:

    • To confirm the presence of PKA in the human placental chorion.
    • To determine the chemical characteristics of placental PKA.
    • To measure PKA activity throughout human pregnancy.

    Main Methods:

    • Assessed PKA activity by measuring ATP production.
    • Determined optimal pH, magnesium ion concentration, and Km for ATP.
    • Quantified specific enzyme activity (pmol ATP/mg protein/minute) in placental samples from each trimester.

    Main Results:

    • Placental PKA exhibits optimal activity between pH 7.5-8.5, with maximal activation at 5mM Mg2+ and a Km for ATP of 4.94 µM.
    • Specific PKA activities were 44.67 ± 5.29 (1st trimester), 45.24 ± 9.13 (2nd trimester), and 34.19 ± 4.95 (3rd trimester).
    • A statistically significant decrease in PKA activity was observed in the third trimester compared to the first and second trimesters (p < 0.001).

    Conclusions:

    • Cyclic AMP-dependent protein kinase is present in the human placental chorion.
    • Placental PKA activity shows a significant decline during the third trimester of pregnancy.
    • These findings provide baseline biochemical data on PKA in the human placenta across gestation.
