    Summary

    Murine cytomegalovirus (MCMV) infection combined with antilymphocyte globulin (ALG) causes progressive glomerulonephritis and renal failure in mice. This suggests viruses can trigger immune-mediated kidney disease.

    Background:

    • Acute murine cytomegalovirus (MCMV) infection typically causes transient glomerulonephritis.
    • Antilymphocyte globulin (ALG) alone does not induce progressive renal disease.

    Purpose of the Study:

    • To investigate the role of MCMV infection in conjunction with ALG in the development of progressive glomerulonephritis.

    Main Methods:

    • Mice were infected with MCMV and treated with ALG.
    • Kidney pathology was assessed using immunofluorescence and electron microscopy.
    • Proteinuria and azotemia were monitored.

    Main Results:

    • Mice receiving both MCMV and ALG developed severe proteinuria, azotemia, and glomerular crescents.
    • Viral antigens were localized to mesangial zones.
    • Immune deposits (rabbit IgG, mouse IgG, C3) were found along capillary walls.
    • Virus was identified in glomerular mesangial cells.

    Conclusions:

    • A multiphasic mechanism involving persistent MCMV in the mesangium, followed by immune complex deposition, leads to amplified immune-mediated glomerular injury.
    • Viruses localizing in the glomerular mesangium should be considered potential causes of progressive glomerulonephritis.
