    Platelet activation by AGEPC rapidly alters inositol phospholipids. Phosphatidylinositol 4,5-bisphosphate levels decrease within seconds, suggesting its crucial role in early platelet signaling.

    • Platelets play a critical role in hemostasis and thrombosis.
    • Inositol phospholipids are key signaling molecules in cellular processes.
    • AGEPC (1-O-alkyl-2-acetyl-sn-glyceryl-3-phosphorylcholine) is a potent platelet activator.

    Purpose of the Study:

    • To investigate the rapid changes in inositol phospholipids upon AGEPC stimulation of rabbit platelets.
    • To elucidate the role of specific phosphoinositides in the early phase of platelet activation.

    Main Methods:

    • Washed rabbit platelets were labeled with [3H]inositol.
    • Platelets were stimulated with AGEPC (1-O-alkyl-2-acetyl-sn-glyceryl-3-phosphorylcholine) at various concentrations and time points.
    • Changes in radioactivity of phosphatidylinositol 4,5-bisphosphate (TPI), phosphatidylinositol 4-phosphate, and phosphatidylinositol (PI) were measured.

    • A rapid, dose-dependent decrease in [3H]TPI was observed within 5 seconds of AGEPC stimulation.
    • Radioactivity in TPI increased shortly after the initial decrease.

    • LysoGEPC, an inactive analogue, did not induce phosphoinositide changes.

    Conclusions:

    • The rapid and dose-dependent changes in TPI suggest its close association with the initial interaction of AGEPC with platelets.
    • Inositol phospholipid metabolism is rapidly modulated during early platelet activation.
    • TPI is likely involved in the early signaling events triggered by AGEPC.
