Phosphate/hexose 6-phosphate antiport in Streptococcus lactis.

P C Maloney, S V Ambudkar, J Thomas

    Journal of Bacteriology
    |April 1, 1984
    PubMed
    Summary

Streptococcus lactis 7962 exhibits a unique phosphate exchange mechanism, independent of ion gradients. This novel anion antiporter facilitates the exchange of inorganic phosphate with various sugar phosphates.

    Area of Science:

    • Microbiology
    • Molecular Biology
    • Biochemistry

    Background:

    • Inorganic phosphate transport is crucial for bacterial metabolism.
    • Specific transport mechanisms vary significantly between bacterial strains and species.

    Purpose of the Study:

    • To investigate the mechanism of inorganic phosphate transport in Streptococcus lactis 7962.
    • To identify the substrates and characteristics of the phosphate transport system.

    Main Methods:

    • Cellular phosphate uptake and exchange assays.
    • Inhibition studies with various anions and sugar phosphates.
    • Analysis of transport activity in relation to growth conditions.

    Main Results:

    • Streptococcus lactis 7962 demonstrated rapid inorganic phosphate exchange, unlike other tested strains.
    • The exchange was independent of membrane potential and pH gradients.
    • Arsenate and several sugar 6-phosphates were identified as alternative substrates for exchange.
    • Transport activity correlated with specific carbohydrate supplementation during bacterial growth.

    Conclusions:

    • A novel anion antiporter in Streptococcus lactis 7962 mediates the exchange of inorganic phosphate (or arsenate) with sugar 6-phosphates.
    • This transporter plays a role in nutrient uptake and its expression is regulated by growth substrates.
