    Background:

    • The human leukocyte antigen (HLA) system plays a critical role in immune responses and transplantation.
    • Specificities w4 and w6 are genetically linked to the HLA-B locus, but their precise molecular relationship remains to be fully elucidated.

    Purpose of the Study:

    • To investigate the molecular association of human alloantigenic specificities w4 and w6 with known HLA-B locus products.
    • To determine if w4 and w6 represent distinct antigenic determinants on the same or different molecules.

    Main Methods:

    • Solubilization of membrane-bound antigens from the RPMI 4265 lymphoblastoid cell line using papain digestion.
    • Copurification of w4 and w6 specificities with HLA-B7 and HLA-B12.
    • Sequential immunoprecipitation using specific alloantisera and a radiolabeled antigen pool.

    Main Results:

    • HLA-B7 and w6, genetically associated, were identified as distinct determinants on the same molecule.
    • HLA-B12 and w4, also genetically associated, were identified as distinct determinants on a second molecule.
