    Hydrocortisone did not improve islet cryopreservation viability. This study found functional islet survival was approximately 30% after freeze-thawing, regardless of steroid treatment.

    Purpose of the Study:

    • To investigate the potential benefit of hydrocortisone during islet cryopreservation.
    • To assess the functional viability of cryopreserved islets.

    Main Methods:

    • Neonatal murine islets were isolated and cultured.
    • Islets were cryopreserved using a programmed freezing system in RPMI 1640 medium with or without hydrocortisone.
    • Glucose-stimulated insulin release was measured before and after cryopreservation.

    • Fresh islets showed significant glucose-stimulated insulin release (mean 113% above basal).
    • After cryopreservation and thawing, functional islet survival was approximately 30%.
