    Background:

    • Chlamydia trachomatis lymphogranuloma venereum (CLV) is a significant cause of sexually transmitted infections.
    • Accurate and sensitive diagnostic methods are essential for timely treatment and control of CLV.

    Purpose of the Study:

    • To develop and characterize a solid-phase immunoassay for detecting the major outer membrane protein (MOMP) of C. trachomatis LGV serotype L2.
    • To establish the sensitivity and specificity of the developed immunoassay.

    Main Methods:

    • Development of a solid-phase immunoassay utilizing immunoadsorbent-purified rabbit anti-L2 MOMP immunoglobulin G (IgG) as a capture antibody.
    • Conjugation of the IgG antibody to horseradish peroxidase or radioiodination with 125I for use as a detection probe.
    • Passive adsorption of capture antibodies to microtiter plates.

    Main Results:

    • The immunoassay demonstrated high sensitivity, detecting as little as 0.5 to 1 ng of purified L2 MOMP.
    • The assay could detect 5 X 10(3) elementary body inclusion-forming units.
    • Approximately 100 C. trachomatis intracytoplasmic inclusions per assay were detectable.

    Conclusions:

    • A sensitive and specific solid-phase immunoassay for detecting C. trachomatis LGV serotype L2 MOMP has been successfully developed.
    • This assay holds potential for improved diagnosis of CLV infections.
