    Summary

This study reveals that fresh thyroid glands are optimal for examining C-cells, which produce calcitonin (CT). Autopsy samples are unsuitable, and C-cells are most concentrated in the upper-middle thyroid lobe.

    Area of Science:

    • Endocrinology
    • Histology
    • Oncology

    Context:

    • C-cells, the source of calcitonin (CT), are implicated in medullary thyroid carcinoma.
    • The precise morphology and distribution of normal human C-cells remain poorly understood.
    • Immunoperoxidase staining offers high specificity for cellular identification.

    Purpose:

    • To detail the morphology, distribution, and population of C-cells in fresh human thyroid glands.
    • To investigate carcinoembryonic antigen (CEA) activity in C-cells and solid cell nests.
    • To assess the suitability of autopsy thyroid tissue for C-cell analysis.

    Summary:

    • Fresh thyroid glands (n=15) were analyzed using immunoperoxidase staining for C-cells.
    • C-cells were identified in all fresh samples, exhibiting varied morphology and parafollicular distribution.
    • C-cell population density was highest (47-111 cells/section) at the upper-middle thyroid lobe junction.

    Impact:

    • Autopsy thyroids are unsuitable for C-cell examination via immunoperoxidase staining.
    • This research provides a more accurate C-cell population count and distribution map.
    • Findings enhance understanding of C-cell biology and medullary thyroid carcinoma origins.
