    Summary

Researchers identified the full amino acid sequence of insulin-like growth factor I (IGF-I) from human liver cDNA. This reveals IGF-I is synthesized as a precursor protein requiring proteolytic processing.

    Area of Science:

    • Biochemistry
    • Molecular Biology
    • Genetics

    Background:

    • Somatomedins (SM), also known as insulin-like growth factors (IGF), are peptides with significant growth-promoting effects.
    • IGF-I (SM-C) and IGF-II are homologous peptides, with determined amino acid sequences for only these two.
    • The liver is a primary site for IGF production, with other tissues also contributing, especially during fetal development.

    Purpose of the Study:

    • To determine the complete amino acid sequence of human IGF-I.
    • To elucidate the synthesis pathway of IGF-I.

    Main Methods:

    • Analysis of human liver cDNA nucleotide sequence.
    • Deduction of the complete amino acid sequence of IGF-I.

    Main Results:

    Related Experiment Videos

    • The nucleotide sequence of human liver cDNA encoding IGF-I was determined.
    • The deduced amino acid sequence of IGF-I comprises 70 residues.
    • The IGF-I coding region is flanked by sequences encoding amino-terminal and carboxyl-terminal peptides.

    Conclusions:

    • IGF-I is synthesized as a precursor protein.
    • Formation of mature IGF-I involves proteolytic cleavage at both the amino and carboxyl termini of the precursor.
