    Summary

    Cyclosporin A (cyA) generally preserves human polymorphonuclear leukocyte functions like phagocytosis and bacterial killing. However, high concentrations of cyA may slightly impair Staphylococcus aureus killing in certain conditions.

    Background:

    • Polymorphonuclear leukocytes (PMNs) are crucial for innate immunity.
    • Cyclosporin A (cyA) is an immunosuppressive drug used in transplantation.
    • The impact of cyA on PMN function requires detailed investigation.

    Purpose of the Study:

    • To evaluate the effects of cyclosporin A (cyA) on key human polymorphonuclear leukocyte (PMN) functions.
    • To assess cyA's impact on phagocytosis, metabolic burst, bacterial killing, and chemotaxis.
    • To determine if cyA affects PMN function in renal transplant recipients.

    Main Methods:

    • Human PMNs were incubated with varying concentrations of cyA (10 and 50 µg/ml).
    • Phagocytosis, hexose monophosphate shunt activity, bacterial killing (using Pseudomonas aeruginosa and Staphylococcus aureus), and chemotaxis were measured.

    • Cyclosporin A (cyA) generally maintains human polymorphonuclear leukocyte (PMN) functional integrity.
    • High-dose cyA may have a minor, transient effect on Staphylococcus aureus killing.
    • The immunosuppressive effects of cyA do not appear to significantly compromise PMN-mediated immunity in vivo.
