    Summary

Researchers controlled plasmid DNA replication and copy number by regulating the priming promoter. Deleting natural promoters and using inducible promoters demonstrated promoter control over DNA replication.

    Area of Science:

    • Molecular Biology
    • Genetics
    • Biochemistry

    Background:

    • ColE1 plasmids are widely used in molecular biology for DNA cloning and protein expression.
    • Replication of ColE1 plasmids is tightly regulated by RNA I and a specific promoter.
    • Understanding plasmid replication control is crucial for optimizing recombinant DNA technology.

    Purpose of the Study:

    • To investigate the role of the priming promoter in ColE1 plasmid replication.
    • To determine if controlling the priming promoter is sufficient to regulate plasmid copy number.
    • To explore the use of inducible promoters for controlled plasmid replication.

    Main Methods:

    • Construction of ColE1 derivative plasmids with deleted natural replication promoters and RNA I control elements.
    • Insertion of constitutive or inducible (lacUV5) promoters upstream to drive read-through transcription.
    • Utilizing IPTG to regulate lac repressor activity and control plasmid levels in vivo.

    Main Results:

    • Plasmid replication was successfully initiated by read-through transcription from engineered promoters.
    • Controlled plasmid levels were achieved in vivo by regulating the inducible lacUV5 promoter with IPTG.
    • Demonstrated that regulation of the priming promoter alone can control DNA replication.

    Conclusions:

    • The priming promoter plays a critical role in ColE1 plasmid DNA replication initiation.
    • Controlling the activity of the priming promoter is sufficient to determine and regulate plasmid copy number.
    • Inducible promoters offer a powerful tool for achieving controlled plasmid replication in molecular biology applications.
