    Summary

    Aggregated beta 2-microglobulin binds to C1q, a complement protein, but does not activate it. This suggests aggregation state influences complement activation by beta 2-microglobulin.

    Background:

    • Beta 2-microglobulin (B2M) is a protein associated with cell membranes.
    • Amyloidosis involves the aggregation of proteins, including B2M.
    • The complement system, particularly C1q, plays a role in immune responses.

    Purpose of the Study:

    • To investigate the interaction between aggregated beta 2-microglobulin and C1q.
    • To determine if aggregated beta 2-microglobulin can activate the complement system.

    Main Methods:

    • Fluid and solid phase binding assays were used to assess B2M-C1q interaction.
    • Affinity chromatography was employed to study monomeric B2M reactivity with C1q.
    • Complement activation assays were performed using serum.

    Main Results:

    • Aggregated beta 2-microglobulin demonstrated efficient binding to C1q in both fluid and solid phase assays.
    • Monomeric beta 2-microglobulin also showed reactivity with C1q.
    • Despite binding C1q, aggregated beta 2-microglobulin failed to activate the C1 complex in serum.

    Conclusions:

    • Beta 2-microglobulin, in aggregated form, can bind to C1q.
    • The inability of aggregated B2M to activate the C1 complex may be dependent on the specific aggregation method used.
