    Summary

This study demonstrates a modified microchamber method for quantifying granulocyte chemotaxis and chemokinesis using nitrocellulose filters. The new approach offers comparable accuracy to Boyden chambers with improved reproducibility and reduced cell requirements.

    Area of Science:

    • Immunology
    • Cell Biology
    • Biotechnology

    Background:

    • Chemotaxis and chemokinesis are crucial granulocyte functions.
    • Standard Boyden chambers are widely used but have limitations.
    • Alternative methods are needed for specific filter types and reduced cell counts.

    Purpose of the Study:

    • To evaluate a 48-well microchamber adapted for nitrocellulose filters.
    • To compare its performance against standard Boyden chambers for granulocyte migration assays.
    • To assess the accuracy, reproducibility, and cell-input requirements of the modified method.

    Main Methods:

    • Utilized a 48-well microchamber with nitrocellulose filters for granulocyte migration.
    • Compared chemotaxis and chemokinesis quantification to traditional Boyden chambers.
    • Analyzed variability in individual and replicate readings.

    Main Results:

    • The modified microchamber showed accuracy comparable to Boyden chambers.
    • Reproducibility between replicate values was superior to Boyden chambers.
    • Optimal chemoattractant doses were similar across both chamber types.

    Conclusions:

    • The 48-well microchamber with nitrocellulose filters is a viable alternative for studying granulocyte chemotaxis and chemokinesis.
    • This method offers advantages in reproducibility and reduced cell usage, beneficial for pediatric or neutropenic patient samples.
    • It facilitates research requiring cellulose filter compatibility.
