Alveolar macrophage plasminogen activator.

M Schuyler, W Forman

    Experimental Lung Research

    Summary

Rabbit alveolar macrophages secrete plasminogen activator, a key enzyme in inflammation and fibrinolysis. Secretion is significantly enhanced in macrophages activated by bacillus Calmette-Guérin (BCG).

    Area of Science:

    • Immunology
    • Cell Biology
    • Biochemistry

    Background:

    • Plasminogen activator (PA) is a neutral serine protease involved in inflammation and fibrinolysis.
    • Alveolar macrophages play roles in immune responses and tissue homeostasis.
    • Understanding PA secretion by macrophages is crucial for inflammatory and cancer research.

    Purpose of the Study:

    • To investigate the secretion of plasminogen activator by rabbit alveolar macrophages.
    • To determine factors influencing PA secretion, particularly in response to macrophage activation.

    Main Methods:

    • Culturing rabbit alveolar macrophages from normal and BCG-pretreated animals.
    • Measuring PA secretion into culture media.
    • Assessing the effects of protein synthesis inhibitors, concanavalin A, and phorbol myristate acetate on secretion.
    • Analyzing PA molecular weights and correlating secretion with Bordetella bronchiseptica presence.

    Main Results:

    • Rabbit alveolar macrophages secrete PA into the medium, not intracellularly.
    • Secretion is dependent on viable cells, blocked by protein synthesis inhibitors, and enhanced by specific activators.
    • BCG pretreatment significantly increased PA secretion compared to normal macrophages.
    • PA exists in two forms (28,000 and 45,000 molecular weight); Bordetella bronchiseptica presence did not correlate with secretion.

    Conclusions:

    • Rabbit alveolar macrophages secrete a serine protease similar to PA found in mouse peritoneal macrophages.
    • Macrophage activation, particularly by BCG, markedly enhances PA secretion.
    • This enhanced secretion suggests a role for activated alveolar macrophages in inflammatory and fibrinolytic processes.
