    Background:

    • Previous studies suggested antitumor effects from immunoadsorption using protein A from Staphylococcus aureus.
    • Various column designs have been employed for this therapeutic approach.

    Purpose of the Study:

    • To assess the feasibility and safety of continuous plasma immunoadsorption using protein A chemically linked to crystalline silica in patients with advanced malignancies.
    • To evaluate preliminary antitumor responses in patients undergoing this treatment.

    Main Methods:

    • A Phase I clinical trial involving 20 patients with advanced malignancies.
    • Continuous immunoadsorption of plasma over columns containing protein A-linked crystalline silica.
    • Patients received 1-22 treatments per week, with one calculated plasma volume perfused per treatment.

    Main Results:

    • Side effects, including chills, fever, nausea, and hypotension, were common but generally manageable.
    • Modest antitumor responses were observed: 3/7 melanoma patients and 1/3 breast cancer patients showed <50% response.
