Trazodone, an antidepressant, partially reduced cocaine

    Area of Science:

    • Clinical Pharmacology
    • Neuroscience
    • Psychiatry

    Background:

    • Long-term cocaine use is associated with depression and dysphoria.
    • The interaction between antidepressants and cocaine in humans is not well understood.

    Purpose of the Study:

    • To investigate the physiological and subjective effects of cocaine hydrochloride after pretreatment with trazodone hydrochloride or placebo.

    Main Methods:

    • A double-blind study involving eight healthy male cocaine users.
    • Participants received either trazodone hydrochloride (100 mg oral) or placebo.
    • Physiological (blood pressure, pupil size, skin temperature, plasma catecholamines) and subjective effects of a 2-mg/kg oral cocaine hydrochloride dose were measured.

    Main Results:

    Related Experiment Videos

    • Trazodone pretreatment diminished cocaine-induced increases in blood pressure, pupil size, and decreased skin temperature.
    • Trazodone did not alter plasma epinephrine levels post-cocaine but increased the norepinephrine response.
    • Subjective effects: Cocaine-induced euphoria was unaffected, but tension and shakiness were reduced by trazodone.

    Conclusions:

    • Trazodone partially mitigates some physiological and subjective adverse effects of acute cocaine administration in humans.
    • Further research is needed to understand the complex interactions between trazodone and cocaine, particularly regarding catecholamine responses.
