Chemicals and cancer: initiation and promotion

What happens during the latency period in tumorigenesis--the interval between exposure to a carcinogen and development of detectable neoplastic changes? A two-step process has been delineated: initiation, which is apparently irreversible and occurs at any carcinogen dose level, and promotion, which is dose- and time-dependent and thus may be a reversible step in tumor development.
