Measuring IgG anti-A/B titres using dithiothreitol (DTT).

R C Knight

    Journal of Clinical Pathology

    Summary

This study compared two sulphydryl compounds, 2-mercaptoethanol (2ME) and dithiothreitol (DTT), for titrating IgG antibodies. A rapid DTT method showed good correlation with established 2ME and DTT techniques.

    Area of Science:

    • Immunology
    • Biochemistry

    Background:

    • Accurate titration of immunoglobulin G (IgG) antibodies, specifically anti-A and anti-B, is crucial for blood transfusion compatibility.
    • Sulphydryl compounds like 2-mercaptoethanol (2ME) and dithiothreitol (DTT) are used to enhance antibody detection in antiglobulin tests.
    • Established methods often involve multiple stages, potentially increasing processing time.

    Purpose of the Study:

    • To evaluate a rapid, one-stage technique for titrating IgG anti-A and anti-B using dithiothreitol (DTT).
    • To compare the efficacy of this new DTT method against existing two-stage methods employing either 2ME or DTT.

    Main Methods:

    • A one-stage antiglobulin test was developed utilizing DTT for IgG anti-A and anti-B titration.
    • Results from the DTT method were compared against standard two-stage titration methods using 2ME and DTT.

    Main Results:

    • The rapid one-stage DTT titration method demonstrated strong correlation with established two-stage methods.
    • This suggests the new method provides comparable accuracy to existing protocols.

    Conclusions:

    • A rapid one-stage DTT antiglobulin test is a viable and effective method for titrating IgG anti-A and anti-B.
    • This technique offers a potentially faster alternative for antibody quantification in serological testing.
