    Area of Science:

    • Microbiology
    • Molecular Biology
    • Biochemistry

    Background:

    • Plasmid DNA isolation from lactic streptococci can be challenging, especially for large plasmids.
    • Existing methods may not be efficient for preparative or screening purposes.

    Purpose of the Study:

    • To develop a rapid and efficient procedure for isolating large plasmid DNA (>30 megadaltons) from lactic streptococci.
    • To provide a method suitable for both preparative applications and high-throughput screening.

    Main Methods:

    • A novel protocol for the rapid isolation of plasmid DNA from lactic streptococci.
    • Adaptation of the protocol for both preparative scale and scaled-down screening.

    Main Results:

    • The procedure successfully isolates plasmid DNA larger than 30 megadaltons.
    • Sufficient quantities of plasmid DNA were obtained for downstream applications like restriction analysis, cloning, and transformation.
    • The scaled-down protocol effectively screened streptococcal strains, revealing previously undetected large plasmids.

    Conclusions:

    • The described methodology offers a significant advancement in the isolation of large plasmid DNA from lactic streptococci.
    • This technique enhances the ability to study plasmid content and diversity in these bacteria.
