    Background:

    • Autoimmune diseases are characterized by the immune system attacking self-tissues.
    • Immunoglobulin light chains are critical components of antibodies involved in immune responses.
    • Specific mouse models are used to study the pathogenesis of human autoimmune diseases.

    Purpose of the Study:

    • To investigate potential molecular differences in immunoglobulin light chains in autoimmune mouse sera.
    • To identify specific immunoglobulin abnormalities associated with the development of autoimmunity.

    Main Methods:

    • High-resolution two-dimensional gel electrophoresis was employed for serum protein analysis.
    • Silver staining was used for sensitive detection of proteins.
    • Immunoglobulin G (IgG) was removed from sera via absorption with staphylococci.

    Main Results:

    • Unusually acidic immunoglobulin light chains were prominently detected in MRL/Mp-lpr/lpr mice sera.
    • These acidic light chains were also found in other autoimmune-prone strains (NZB/NZW, BXSB) and to a lesser extent in normal mice.
    • The concentration of acidic light chains increased with age in MRL/Mp-lpr/lpr mice, correlating with disease progression.
    • Acidic light chains were integral components of complete IgG molecules and were found in cryoglobulins and glomerular eluates.

    Conclusions:

    • Acidic immunoglobulin light chains are a potential biomarker for certain autoimmune conditions in mice.
    • These aberrant light chains may play a role in the formation of immune complexes.
    • The findings suggest a link between specific immunoglobulin light chain modifications and autoimmune pathogenesis.
