    Background:

    • Hemophilia A and von Willebrand's disease are bleeding disorders requiring effective factor VIII concentrates.
    • Existing treatments may have limitations in purity and efficacy.
    • Development of highly purified factor VIII is crucial for improved patient outcomes.

    Purpose of the Study:

    • To develop and characterize two high-purity factor VIII concentrates (Type I and Type II).
    • To evaluate the efficacy of these concentrates in preclinical models.
    • To assess the purification process and specific activities of the factor VIII preparations.

    Main Methods:

    • Cryoprecipitation of fresh frozen plasma.
    • Selective precipitation of fibrinogen using glycine buffer.
    • Factor VIII precipitation with sodium chloride and further purification by gel filtration (Type II).
    • Analysis of factor VIII procoagulant activity (VIII:C) and von Willebrand factor (VIIIR:RC) recovery and specific activity.

    Main Results:

    • Type I and Type II factor VIII concentrates demonstrated high recovery rates for both VIII:C and VIIIR:RC.
    • Type II preparations exhibited significantly higher specific activities for both VIII:C and VIIIR:RC compared to Type I.
    • Purified Type II concentrate was free of fibrinogen and fibronectin, showing potent correction of bleeding times in von Willebrand's disease models.

    Conclusions:

    • The developed Type I and Type II factor VIII concentrates are highly pure and possess significant procoagulant and von Willebrand factor activities.
    • The Type II preparation, further purified by gel filtration, shows superior specific activity and potential for treating hemophilia A and von Willebrand's disease.
    • These findings support the clinical trial readiness of these novel factor VIII concentrates.
