[Detection of acid mucopolysaccharides by hematoxylin]

Author studied the effect of the sequence of the dissolution of components of alumhaematoxylin solution with potassium iodate in it and the effect of the quantity of components on the staining capacity of the dye. Potassium iodate should be added after haematoxylin and alum are dissolved, otherwise the staining of the cartilage and mucus will be less successful. The best staining effect of tissue specimens containing acid mucopolysaccharides and cell nuclei as well can be achieved by the use of haematoxylin solution containing 0,5 per cent of alum. Sections prepared by the freezing technique should be stained by haematoxylin containing 5 per cent of alum.
