    Background:

    • Elevated immunoglobulin E (IgE) levels are characteristic of atopic diseases.
    • Monocytes play a complex role in immune regulation and inflammation.

    Purpose of the Study:

    • To investigate the role of monocytes in regulating IgE biosynthesis in asthmatic children.
    • To identify potential mechanisms contributing to heightened IgE production in atopic individuals.

    Main Methods:

    • Separation of monocytes from mononuclear cells using adherence.
    • Activation of monocytes with lipopolysaccharide (LPS).
    • Measurement of IgE production in patient and normal cell cultures.

    Main Results:

    • Mononuclear cells (MNCs) from asthmatic children produced significantly more IgE than those from healthy controls.
    • Depletion of monocytes reduced IgE production in asthmatic MNCs but not in normal MNCs.
    • Patient monocytes enhanced T cell activity related to IgE biosynthesis in both groups.

    Conclusions:

    • Suppressor adherent cells in allergic patients inhibit regulatory T cell function.
    • These suppressor cells may contribute to the pathogenesis of increased IgE production in atopy.
