    Summary

    Background:

    • Phencyclidine (PCP) is a dissociative anesthetic with known psychotomimetic effects.
    • Understanding the neurobehavioral effects of PCP is crucial for its clinical and forensic applications.

    Purpose of the Study:

    • To investigate the effects of phencyclidine (PCP) on tactile startle and behavioral habituation in male rats.
    • To determine the dose-response relationship and time course of PCP's effects on these behaviors.

    Main Methods:

    • Male rats received subcutaneous injections of saline or varying doses of PCP (0.25–5.0 mg/kg).
    • Animals were exposed to 121 tactile (air-puff) stimuli at 15-second intervals.
    • Startle responses and behavioral habituation rates were measured.

    Main Results:

    • PCP increased startle responses in an inverted-U dose-response manner, with maximal effect at 1.0 mg/kg.
    • Lower doses of PCP (0.5 or 1.0 mg/kg) reduced the rate of behavioral habituation.
    • This habituation impairment was independent of the time elapsed since PCP administration.

    Conclusions:

    • Impaired behavioral habituation appears to be the threshold effect of PCP on tactile startle.
    • These findings contribute to understanding the neurobiological mechanisms underlying PCP's behavioral effects.
