    Acute hypertension transiently alters the surface charge of cerebral arterioles, affecting their permeability. This study used cationized ferritin to visualize changes in anionic groups on endothelial cells during hypertension.

    Background:

    • Cerebral arteriolar endothelium possesses anionic groups that influence vascular permeability.
    • Hypertension can alter the blood-brain barrier, but the underlying endothelial changes are not fully understood.

    Purpose of the Study:

    • To investigate alterations in endothelial anionic groups during angiotensin-induced acute hypertension.
    • To correlate changes in surface charge with increased cerebral arteriolar permeability.

    Main Methods:

    • Localization of anionic groups using cationized ferritin (CF) on fixed and live cerebral arterioles.
    • Assessment of vascular permeability using horseradish peroxidase (HRP) in normotensive and hypertensive animal models.
    • Microscopic analysis of CF distribution on endothelial cells and in vesicles.

    Main Results:

    • Normotensive arterioles showed uniform CF binding on the luminal surface.
    • Hypertensive arterioles with increased permeability to HRP exhibited reduced or lost CF binding, indicating altered anionic charge.

    Conclusions:

    • Increased cerebral arteriolar permeability during acute hypertension is associated with a transient alteration of endothelial surface charge.
    • The findings highlight a dynamic change in endothelial glycocalyx during hypertensive events.
    • Further research is needed to elucidate the precise mechanism behind the surface charge alteration.
