    Summary

    Mitoxantrone showed limited efficacy in advanced cancers, particularly in patients previously treated with adriamycin. The anthraquinone caused significant leukopenia and thrombocytopenia in most patients.

    Background:

    • Mitoxantrone is an anthraquinone chemotherapy agent.
    • Advanced solid tumors and malignant lymphomas often exhibit resistance to standard chemotherapies.

    Purpose of the Study:

    • To evaluate the efficacy and toxicity of mitoxantrone in patients with advanced solid tumors and lymphomas.
    • To assess response rates in breast cancer patients with and without prior adriamycin exposure.

    Main Methods:

    • A phase II clinical trial involving 31 patients with advanced solid tumors and 2 with malignant lymphomas.
    • Mitoxantrone administered via a 5-day schedule (2-4 mg/m2) or a 1-day schedule (6-12 mg/m2), repeated every 4 weeks.
    • Evaluation of response in 18 evaluable breast cancer patients.

    • Significant myelosuppression (leukopenia, thrombocytopenia) is a major toxicity of mitoxantrone.
    • Further investigation may be warranted in specific patient populations or treatment settings.
