    Summary

    Toxic doses of methaqualone impair human blood platelet function, inhibiting aggregation responses to adenosine diphosphate (ADP) and collagen. This platelet dysfunction may explain methaqualone-induced bleeding tendencies.

    Background:

    • Methaqualone overdose is associated with a bleeding tendency.
    • The underlying mechanism of this bleeding risk is not fully understood.

    Purpose of the Study:

    • To investigate the effects of methaqualone on human blood platelet function.
    • To determine if methaqualone-induced platelet dysfunction contributes to bleeding complications.

    • In vitro studies exposed human platelets to varying concentrations of methaqualone and diphenhydramine.
    • Platelet aggregation was measured in response to adenosine diphosphate (ADP) and collagen.
    • In vivo studies assessed platelet aggregation in rabbits after methaqualone administration.
