Cimetidine does not alter ibuprofen kinetics after a single dose.

K A Conrad, M Mayersohn, M Bliss

    British Journal of Clinical Pharmacology
    |October 1, 1984
    PubMed
    Summary

Cimetidine did not affect how quickly the body processed a single dose of ibuprofen in healthy men. Therefore, adjusting ibuprofen dosage is unnecessary when taken with cimetidine.

    Area of Science:

    • Pharmacology
    • Drug Metabolism
    • Clinical Pharmacology

    Background:

    • Ibuprofen is a common nonsteroidal anti-inflammatory drug (NSAID).
    • Cimetidine is a histamine H2 receptor antagonist used to reduce stomach acid.
    • Potential drug interactions between ibuprofen and cimetidine warrant investigation.

    Purpose of the Study:

    • To determine if cimetidine alters the serum elimination rate of ibuprofen.
    • To assess the clinical significance of co-administering cimetidine and ibuprofen.

    Main Methods:

    • A single dose of ibuprofen was administered to healthy male volunteers.
    • Serum ibuprofen levels were monitored over time.
    • The pharmacokinetic profile of ibuprofen was analyzed with and without co-administered cimetidine.

    Main Results:

    • Cimetidine did not significantly affect the disappearance rate of ibuprofen from serum.
    • No statistically significant differences were observed in key pharmacokinetic parameters of ibuprofen.

    Conclusions:

    • Cimetidine does not inhibit the metabolism or elimination of a single dose of ibuprofen.
    • Current ibuprofen dosing regimens are likely safe and effective when co-administered with cimetidine in healthy individuals.
