    Background:

    • Kupffer cells are the primary resident macrophages in the liver.
    • Their roles as accessory and antigen-presenting cells in liver immunity require further elucidation.

    Purpose of the Study:

    • To investigate the functional capabilities of Kupffer cells in immune responses.
    • To determine if Kupffer cells can present liver-specific protein (LSP) and act as accessory cells.

    Main Methods:

    • Sprague-Dawley rats were immunized with LSP.
    • Kupffer cells were isolated and their phagocytic activity assessed.
    • Lymphocyte proliferation assays were performed with and without Kupffer cells.
    • In vitro stimulation assays were conducted using LSP, lymphocytes, and Kupffer cells.

    Main Results:

    • Over 70% of isolated Kupffer cells exhibited phagocytic activity.
    • Macrophage depletion abolished concanavalin A-induced lymphocyte proliferation, which was restored by Kupffer cells.
    • In vitro stimulation with LSP led to lymphocyte binding to Kupffer cells.
    • Enhanced 3H-thymidine incorporation was observed when lymphocytes were incubated with Kupffer cells from immunized rats.

    Conclusions:

    • Kupffer cells possess significant phagocytic capabilities.
    • Kupffer cells effectively function as accessory cells, restoring lymphocyte proliferation.
    • Kupffer cells act as potent antigen-presenting cells for liver-specific protein (LSP).
