    Background:

    • Cyclophosphamide is a known teratogen.
    • Understanding its effects on limb development is crucial for risk assessment.

    Purpose of the Study:

    • To investigate the teratogenic effects of cyclophosphamide on rat forelimb development.
    • To characterize the skeletal malformations induced by cyclophosphamide exposure.

    Main Methods:

    • Pregnant rats received a single dose of cyclophosphamide (20 mg/kg) on gestation Day 12.
    • Fetuses were collected between gestation Days 18-20.
    • Gross examination, Alizarin red-s and Alcian blue staining, and histochemical analysis were performed.

    Main Results:

    • Observed gross forelimb malformations including micromelia, oligodactyly, brachydactyly, and adactyly.
    • Skeletal analysis revealed postaxial deformities.
    • Histochemistry showed delayed ossification of the radius and a completely unossified ulna, with the ulna being more severely affected.

    • Cyclophosphamide exposure during gestation induces significant forelimb skeletal malformations in rats.
