    Summary

    Chronic L-dopa and carbidopa treatment does not alter whole-body decarboxylation. This suggests that L-dopa’s on-off fluctuations are not due to chronic changes in L-aromatic amino acid decarboxylase activity.

    Background:

    • Levodopa (L-dopa) is a primary treatment for Parkinson's disease.
    • Patients often experience motor fluctuations (on-off phenomena) despite chronic L-dopa therapy.
    • The role of L-aromatic amino acid decarboxylase (AADC) in these fluctuations is not fully understood.

    Purpose of the Study:

    • To investigate the effect of chronic L-dopa and carbidopa treatment on whole-body L-dopa decarboxylation.
    • To determine if chronic changes in AADC activity contribute to L-dopa's on-off fluctuations.
    • To evaluate the mechanism by which carbidopa enhances L-dopa's clinical efficacy.
