Therapeutic concentration of coumarin and predicted dosage regimens

Abstract:
The concentrations of coumarin (C) and its metabolites 7-hydroxy-coumarin (7-OHC) and 7-hydroxycoumarin glucuronide (7-OHCG) were determined in 24 episiotomy patients at the time clinical effectiveness was observed. The patients were on 7:00 h-13:00 h-19:00 h dosage regimen of 2 controlled release tablets (Venalot Depot; 15 mg C per tablet). The C concentrations at the end of the 7:00 h dose was 0.011 +/- 0.0027 micrograms/ml +/- SEM, and 0.036 +/- 0.0057 micrograms/ml +/- SEM 1/2 h after the following dose. The mean therapeutic concentration was about 0.02 micrograms/ml. Based on previously established pharmacokinetic parameters of C in man dosage regimens for i.v. infusion and i.v. push were calculated.
